13 nữ tù nhân xoay quanh những mảnh đời bất hạnh của 13 người đàn bà vì những lý do khách nhau mà bỗng chốc trở thành kẻ tù tội. Gần 30 tập phim được kết cấu từ 13 câu chuyện độc lập với dàn diễn viên riêng biệt. Sự xuất hiện của ca sĩ Thanh Lam và người mẫu Doãn Tuấn thuộc những tập phim cuối cùng của bộ phim truyền hình này. Có kinh nghiệm diễn xuất qua các phim Chuyện của Pao và Đi tìm hạnh phúc, Doãn Tuấn đã thể hiện rất tròn vai bên cạnh người bạn diễn tên tuổi: diva âm nhạc Thanh Lam.

Nội dung tập phim kể về cuộc đời bi thương của cô sinh viên năm cuối trường mỹ thuật. Cô gái sống cùng người mẹ đã không gần đàn ông trong khoảng 20 năm trời (vai diễn của ca sĩ Thanh Lam). Cô giành được tình cảm yêu thương chân thành từ người thầy giáo trẻ tuổi (do nam người mẫu Doãn Tuấn thủ vai) của mình.

Do không biết mặt người yêu của con gái, trong một lần tình cờ được người thầy giáo - lúc đó đang là người yêu của cô con gái nhỏ - giúp đỡ trong cơn hoạn nạn, người đàn bà 40 tuổi này đã có cơ hội tiếp cận và gần gũi với chàng thanh niên kém bà hơn cả chục tuổi. Những xúc cảm hồi xuân trỗi dậy của người mẹ đã gây sự hiểu lầm đến cô con gái tuổi đôi mươi.

Cô bỏ nhà ra đi, vướng vào cạm bẫy tình với một người đàn ông đã có vợ - nhưng người vợ này tâm thần không ổn định. Một lần người vợ bắt gặp cảnh gần gũi của cô gái với chồng, người vợ đã dùng súng bắn "tình địch trẻ", kết cục viên đạn lại cắm sâu vào lồng ngực của chồng. Trong cuộc giằng co khẩu súng giữa người vợ tâm thần không ổn định với cô gái, súng cướp cò mang đi cuộc sống của người vợ kia. Cô gái trở thành kẻ tù tội...
